5-Amino-1MQ is a small-molecule selective inhibitor of nicotinamide N-methyltransferase (NNMT), an enzyme overexpressed in adipose tissue of obese individuals. NNMT consumes SAM and NAD-precursor methyl donors to produce 1-methylnicotinamide, and elevated NNMT activity is associated with reduced adipocyte energy expenditure and impaired NAD/methylation pools. Inhibition of NNMT in preclinical models shrinks adipocytes, increases cellular NAD, and restores methyl-donor availability.
