PNC-27

PNC27

Think of it as a guided missile for rogue cells.

PNC-27 is a lab-made peptide that homes in on cancer cells like a heat-seeking missile. It targets a specific protein that's found on the surface of many cancer cells but not on healthy ones. Once it locks on, PNC-27 forms tiny holes in the cancer cell membranes, causing them to break apart and die.

In lab tests, PNC-27 has shown promise in taking down different types of cancer cells, including those from leukemia and breast cancer. It works by sparing healthy cells, which could mean fewer side effects if it ever makes it to human trials. However, it's important to note that PNC-27 is still in the early stages of research and hasn't been tested in humans yet.

Mechanism of Action

PNC-27 is a synthetic chimeric peptide combining residues 12-26 of the p53 protein (the HDM-2 binding region) with a membrane-penetrating leader sequence derived from penetratin. Preclinical research reports that PNC-27 binds HDM-2 that is aberrantly expressed on the surface membrane of cancer cells, forming transmembrane pores that trigger necrotic cell death selectively in malignant cells while sparing normal cells that do not express membrane HDM-2. PNC-27 is strictly a preclinical research compound with no human clinical efficacy data and no FDA approval for any indication.

Researched Benefits

Selective tumor-cell membranolysis (preclinical)

In-vitro studies across multiple human cancer cell lines (leukemia, melanoma, pancreatic, breast) report selective necrosis of malignant cells versus paired normal controls.

Xenograft tumor reduction (preclinical)

Mouse xenograft models of pancreatic and acute myeloid leukemia report tumor regression or growth delay with systemic PNC-27 administration.

No validated human dosing exists. All published work is in cell culture and rodent models. This is a preclinical research compound; PNC-27 has not been tested in human clinical trials and should not be used in humans for any therapeutic purpose.

Reported Side Effects

  • Human safety profile is unknown — no clinical trial data exist
  • Potential for off-target membrane effects in tissues that transiently express HDM-2
  • Immunogenicity risk from chimeric peptide (theoretical)
  • Injection-site effects (animal data)

Contraindications

  • All human use outside a regulated preclinical/IRB-approved investigational setting
  • Pregnancy and lactation
  • Pediatric use
  • Any patient with active medical conditions — no safety data exist
