PNC-27
PNC27
Think of it as an investigational peptide studied in cancer-cell research.
PNC-27 is a lab-made peptide studied in cell-culture cancer research. It has not been tested in humans, and nothing here describes a treatment. In laboratory studies it appears to target a protein found on the surface of certain cancer cells but not on healthy ones, and in those studies it has been observed to disrupt the cancer-cell membrane.
In cell-based experiments, researchers have reported activity against several cancer cell lines, including leukemia and breast cancer models, with limited effect on healthy cells in the same tests. These are early preclinical findings only. PNC-27 remains an investigational research compound and has not been evaluated for safety or efficacy in people.

Mechanism of Action
PNC-27 is a synthetic chimeric peptide combining residues 12-26 of the p53 protein (the HDM-2 binding region) with a membrane-penetrating leader sequence derived from penetratin. Preclinical research reports that PNC-27 binds HDM-2 that is aberrantly expressed on the surface membrane of cancer cells, forming transmembrane pores that trigger necrotic cell death selectively in malignant cells while sparing normal cells that do not express membrane HDM-2. PNC-27 is strictly a preclinical research compound with no human clinical efficacy data and no FDA approval for any indication.
Researched Benefits
Selective tumor-cell membranolysis (preclinical)
In-vitro studies across multiple human cancer cell lines (leukemia, melanoma, pancreatic, breast) report selective necrosis of malignant cells versus paired normal controls.

- [Sarafraz-Yazdi et al. 2010]
Xenograft tumor reduction (preclinical)
Mouse xenograft models of pancreatic and acute myeloid leukemia report tumor regression or growth delay with systemic PNC-27 administration.
